Instrumentation

MALDI Imaging can be performed with the axial MALDI-TOF instruments rapifleX MALDI Tissuetyperautoflex speed and ultrafleXtreme and with the solariX XR FTMS. All instruments are equipped with the unique smartbeam laser for soft ionization and maximum compatibility with histology.

Tissue

MALDI Imaging was originally developed on fresh frozen tissue, but the choice of tissues has increased in recent years. For example, the measurement of intact proteins is possible from frozen tissue and from certain non-formalin paraffin embedded protocols. Formalin fixed paraffin embedded tissue is amenable for protein analysis after an antigen retrieval process followed by a spatially resolved tryptic digestion. In this case the tryptic peptides, rather than intact proteins, are measured. Many tissue types such as mammalian, insect, or plant tissues have been shown to be accessible by MALDI Imaging.

 

 

Software

flexImaging is the software that features user-friendly acquisition and interpretation of the MALDI Imaging data. It displays the distribution of individual and multiple masses. flexImaging provides a convenient comparison of different datasets, enables molecular histology with the integration of virtual microscopic slides. Datasets can be segmented by hierarchical clustering. Different normalization options are available. SCiLS lab is a statistical software package that features advanced calculations, visualizations, and the analysis of cohorts in biomarker discovery studies.

 

Sample preparation

For MALDI Imaging, sample preparation is key for producing high quality images.  Bruker offers a choice of industry-leading sample aerosol preparation devices that can meet routine needs for imaging multiple classes of analytes but with the flexibility to image challenging research samples.

 

 

Histology

Prior to MALDI Imaging, an optical image measurement of the unprepared tissue section is acquired. This optical image is registered in the instrument and provides the reference for the measurement. After the measurement is completed, the tissue can be stained with conventional histological stains. A virtual microscopic slide of the stained section is then co-registered to the primary image. This links an unambiguous assignment of the mass spectra to the histology. Bruker’s flexImaging software allows you to overlay virtual microscopic slides in full optical resolution with the mass spectrometric information for a detailed understanding of the results.
