The crystal structure of Prolyl Hydroxylase (PHD2) with an iso-quinoline inhibitor (compound A) was elucidated using data collected on a Bruker X8 PROTEUM diffraction system, on a MICROSTAR-H microfocus rotating anode generator equipped with HELIOS™ multilayer graded optics.

Data were measured in 16 hours using a combination of phi and omega scans at different omega and chi positions. The sample diffracted up to a resolution of 1.80 Å and data were used to solve the structure using SAD phasing. The Fe and I ions were easily located in the difference Fourier maps. It was possible to locate 9 S atoms and these were used to improve the phasing.
