Sierra SPR-32- Combining Throughput, Flexibility and Controls

From initial screening to detailed kinetic characterization and thermodynamics measurements, the Sierra SPR-32 system enables high-throughput surface plasmon resonance (SPR) analysis of molecular interactions. Equipped with 32 individually addressable detection spots, this highly sensitive real-time, label-free analytical system is uniquely configured to provide active and control binding data for small molecules, fragments, proteins, peptides, antibodies and nucleic acids. 

Sierra SPR-32 Delivers Throughput with Performance

While capable of testing a single sample at a time, the Sierra SPR-32 system is a robust instrument designed for high-throughput applications such as epitope binning or mapping and antibody, fragment or small molecule screening. Simultaneous injection of up to eight samples facilitates high-throughput assay development and optimization, as well as rapid quantitative analysis of both crude and purified samples.

Assay Controls Increase Confidence in Results

Four independently addressable detection spots are configured in each of the eight channels of the Sierra SPR-32 system. The four-sensor per channel design provides maximum flexibility for control analysis, while maintaining throughput for even the most complex analyses. Samples can be simultaneously tested on up to three control surfaces as well as the active surface. In many SPR applications, early detection of nonspecific binding to control proteins is critical. In small molecule screening and characterization, binding to serum proteins such as HSA or AGP is an integral part of drug development. The additional in-channel control not only halves the number of assay cycles required but also improves data quality by eliminating potential variations between multiple sample preparations.

Flexibility for Optimal Productivity

The 32 detection spots in the Sierra SPR-32 system are addressed by the use of 8 injection needles.  When using the Individual Needle Control (INC) feature, researchers can choose any combination from 1 to 8 needles.  INC optimizes the performance, flexibility and scope of applications possible, while saving time and materials when throughput is not the main requirement.

Up to four buffers can be used simultaneously in the instrument.  The ability to evaluate different pH‘s, ionic strength, detergent or solvent concentration and the effect of co-factors or inhibitors in a single assay yields simpler, optimized assays that ultimately increase throughput and productivity.
