Fast Intuitive Structure Determination II: Crystal Indexing and Data Collection Strategies

Picking up from Fast Intuitive Structure Determination I: Crystal Selection and Mounting, this webinar continues our series presenting single-crystal small-molecule structure determination. In this one-hour webinar, we present skills and techniques for crystal indexing and data collection strategies. Reliable indexing allows for efficient, effective data collection strategies that achieve coverage and redundancy in a minimum period of time. Indexing requires a proper array of reflections, an array that can be built from tools available in APEX3. These tools can be used automatically or interactively to select reflections and sort them into one or multiple arrays to facilitate indexing routine crystals, twins with two or more domains, and modulated structures. As part of the indexing process, we examine the diffraction behavior of the sample to determine data collection parameters such as scan speed, scan width, and maximum resolution. Using this information, and the indexing solution, we can determine a strategy for data collection that will yield highest data quality and efficient instrument utilization.
