Collision energy stepping  (QTOF) for full Glycopeptide Identification

This approach provides and complete and simple solution for glycopeptide identification. It includes:


  • Acquisition of QTOF CID data using  two dedicated collision energies. Those are set to specifically fragment the glycan or the peptide part of the glycopeptide in an automatic way. A highly suitable QTOF for this workflow is e.g. the impact II. Make use of the high glycopeptide sensitivity using the Captive Spray nanoBooster for high glycopeptide sensitivity, which improves ionization efficiency.
  • ProteinScape the bioinformatic platform with the integrated search engine GlycoQuest
    • Glycopeptide spectra classification: ProteinScape searches for characteristic patterns in the MS/MS spectra. The peptide moiety masses of glycopeptides are determined simultaneously. Only glycopeptide spectra are submitted to the GlycoQuest search for glycan identification.
    • Combined glycopeptide table:  After the protein and glycan database search, these search results are combined to one glycoprotein table, where glycan and peptide information is linked.
    • The Spectrum viewer: Allows the fast and simple verification of the ID results for complete glycopeptides. 

ProteinScape supports the analysis of glycopeptide data from other instruments as well.
