Key attributes of the Toxtyper 2.0:

  • Unique and comprehensive library of over 980 toxicology relevant compounds with retention time, MS, MS², and MS³ spectral info
  • Walk up system with ready-to-use LC-MSn methods and push button user interface
  • Simultaneous identification of drugs and metabolites in high and low abundance with sensitivities in the low ng/ml range
  • Value and speed with capabilities for screening, confirmation and semi-quantitation in a single LC-MSn  analysis
  • “Open toxicology“ concept for rapid update of libraries for emerging new drugs and toxins

Ultra high performance liquid chromatography (UHPLC) combined with true MSn and comprehensive drug libraries

Toxtyper 2.0: a complete solution for Clinical Research and Forensic Toxicology

The high demand in clinical research and forensic toxicology for comprehensive, specific, and transferable techniques has led to new developments in the field of Liquid Chromatography-tandem Mass Spectrometry (LC-MS). LC-MSn, combined with library searching, is one of the most promising screening solutions for modern toxicology research.

Libraries of Drugs

The Toxtyper 2.0 holds a high quality spectral library of more than 980 compounds of toxicology research interest with ˜ 3,000 spectra of MS, MS² and MS³ experiments and a smaller targeted library for Drugs of Abuse screening research. The library search module, as part of Brukers' new Toxtyper 2.0 software package, allows for unambiguous and fast (11-minute duty cycle) drug identification. In addition, the Toxtyper supports the Maurer/Wissenbach/Weber LC-MSn library* containing > 4500 drugs, metabolites, and artifacts.

Tailor-made sub-libraries for increased sensitivity

The Toxtyper provides the option to generate targeted libraries for specific needs.

Concentration of substances in whole blood
Example of selected drug detection in whole blood at a concentration of 0,5 ng/ml.

Sample preparation for a wide range of analytes

Different well-established sample preparation methods, such as liquid-liquid extraction (LLE), solid-phase extraction (SPE) or simple protein precipitation, can be used within the Toxtyper workflow and are well-suited for the extraction of a wide range of analytes such as hypnotics, neuroleptics, antidepressants, and many others.

Full Transferability by SmartFrag™

The Toxtyper 2.0 solution is setting new standards in LC-MS/MS toxicology research and transferability from lab-to-lab. Bruker's patented SmartFrag technology virtually removes variation and tuning from the MS/MS process. Furthermore, results and libraries are easily transferable from lab-to-lab and from experiment-to-experiment.

Toxtyper 2.0 complete application solutions

From comprehensive screening to targeted libraries and semi-quantification

Example data demonstrating simultaneous, highly sensitive detection of drugs in human urine. Following simple sample preparation (liquid-liquid-extraction) of 1 mL of urine, the Toxtyper 2.0 permits detection down to 0.8 ng/ml.

Open library concept

The appearance of new drugs, such as synthetic cannabinoids, makes it necessary to update the screening methods quickly. Toxtyper 2.0 provides ready-to-use spectral reference libraries, constantly updated through contributions from our customers and collaboration partners. User-defined library entries can be created in a few mouse clicks, allowing laboratories to quickly respond to new patterns of illicit and licit drug misuse.

Interactivity provides easy access to high-value data

The intuitive and highly dynamic Toxtyper 2.0 graphical user interface can be customized to the users' preference. For easy and fast result evaluation identified compounds can be sorted by various parameters, e.g. by peak intensity, retention time or alphabetically. In addition color coding of deviating parameters in retention time or mass shift, or of lower purity scores, supports the evaluation options.

Screening, confirmation, and semi-quantitation in one run

The standard workflows can be extended by additional automated methods for semi-quantitation using any of the Toxtyper 2.0 libraries. A one-point calibration for the target compounds using internal standards provides data regarding the concentration range (e.g. above or below a target value) within the same single LC-MSn analysis.

Toxtyper Case Study: Analysis of drugs and toxins in the Katharinenhospital Stuttgart

Although drug analysis using immunological methods is fast, it is not always reliable. The available screening tests can easily lead to false positive results. For the confirmation of such immunoassay results,
complex and time-consuming GC-MS analysis has been so far performed by the Stuttgart hospital.

Since March 2017, the Toxtyper system has been used for the screening of drugs and toxins in blood, oral fluid and urine. Without lengthy sample preparation, Toxtyper delivers fast and reliable results that are highly specific and reproducible. Additionally, the Toxtyper achieves a high level of confidence with toxicological analysis before neurological diagnosis of irreversible brain failure.

Case Study

A 34 year old patient with dizziness, nausea and vomiting is admitted to the emergency room. From the anamnesis there is a suspicion of drug consumption, however the patient denies the use of illegal drugs. Urine screening using various immunoassays was negative for Amphetamines, Barbiturates, Benzodiapezepins, Cocaine, Opiates and Cannabis. The Toxtyper identified Mitragynin and several metabolites of Paynanthein and Speciociliantin, which are alkaloids present in leaves and fruits of the Kratom tree. Kratom is used for substitution of Opioids and is not regulated by the German narcotics law.
