Screening and Epitope Mapping

Though modern biotechnologies can produce large libraries of potential binders, the subsequent selection process should be guided on well founded information to ensure an efficient further development.

The Bruker SPR-solutions offers a versatile high-throughput platform for biologics screening. The sensor array allows to screen a target protein against up to 31 different biologics captured on the surface. The robust and valve-less microfluidics design allows to capture proteins from all types of sample origins like from cell supernatants. In addition, the open platform of Bruker’s SPR allows to easily integrate the system in automated processes and scheduling environments, thus increasing the throughput by another dimension.

SPR provides the required data-rich information in a screening. A single injection gives insights into specificity, affinity and kinetics. This information aids in selecting the candidates with the optimal profile such as slowly dissociating antibodies of high specificity.

Besides selecting the most affine and specific hit, novelty in binding mode and epitope are of equal relevance in the selection process. The Bruker SPR-solutions provide the possibility to investigate binding at various binding conditions like pH. Moreover, epitope mapping with up to 24 proteins in a 24x24-format with different experimental set-ups is possible.
