Introduction To TopSpin For Solid State NMR

This 4 day course is designed to introduce users to the Topspin software, provide a greater understanding of the many features within the software, and practical experience at the spectrometer setting up basic Solid State NMR experiments for spin ½ nuclei. 

The course will be taught focusing on TopSpin 4 software, but the majority of concepts are backwards compatible to older versions of TopSpin.

Lectures will be followed by a laboratory session in front of a spectrometer for hands on experience. 

The lecture portion of the course is done in conjunction with the liquids Introduction to TopSpin

Topics and Experiments:

  • Basic sample preparation and rotor packing 
  • Setting up a CPMAS probe:
    • Setting magic angle
    • Pulse calibration
    • Shimming
    • Optimizing Hartmann-Hahn match condition
  • Standard 1-Dimentional 13C CPMAS experiments and various spectral editing sequences
    • Spinning sideband suppression (TOSS)
    • Dipolar dephasing with and without TOSS
    • CPPI for signal assignment
  • Setup CPMAS for different Nuclei (15N 29SI or 31P)
  • 2-Dimentional structure elucidation experiments (FSLG Hetcor)
  • Processing (widow function, FT, phasing, baseline correction)
  • Post Processing (peak picking, integration)
  • Presenting the data (saving/plotting spectra)
  • Quantitative NMR (qNMR)
  • Introduction to automation
  • Computer / TopSpin administration
  • Variable Temperature

Prerequisite:

An understanding of basic NMR concepts is strongly recommended for this course.
