Quantification and application of CEST signals in rodent brain

Dr. Xiaoyong Zhang from the Zhangjiang International Brain Imaging Center (ZIC) at Fudan University starts his talk with an overview of the research goals of the ZIC, namely a focus on method development, the understanding of brain cognitive mechanisms and brain diseases, the development of artificial intelligence, and the development of a multiscale and multimodal brain science database.


Dr. Zhang then elaborates on his contributions to these goals using his BioSpec 11.7 T, focusing on CEST quantification, which he uses for early stroke detection and in combination with pH guided surgeries. After an introduction to CEST and the Z-spectrum, he discusses quantification challenges, such as asymmetric pools on both sides, that direct saturation and solid MT do not add up linearly to the signal, that it is difficult to separate close/overlapping pools, and that r1 contamination is not considered. He then describes his pH guided brain tumor surgery using CEST mapping and his pH responsive Raman probe.


Dr. Zhang details how his multi-pool Lorentzian fitting, which avoids use of reference signals combined with AREX fitting, which takes non-linear fitting and T1 correction into consideration allowed detection of a new NOE signal at 1.6 ppm which can be used for early stoke detection and in glioma heterogeneity in CEST images. He then explains how gliomas could be completely surgically removed using CEST mapping and the pH responsive Raman probe, and that in comparison to a glioma removed via white light guided surgery, no reoccurrence of the tumor could be seen at days 7 to 15.
