timsTOF SCP - High sensitivity for unprecedented depth in immunopeptidomics

Immunopeptides are located on major histocompatibility complex (MHC) molecules. They help the immune system to recognize foreign substances and combat infections. Due to their low abundances in general and a non-tryptic nature, high-complexity and diversity, LC-MS analysis poses special challenges.

The timsTOF SCP allows for in depth analysis of complex mixtures with ultra-high sensitivity. In addition, PASEF® powered by trapped ion mobility (4D-Proteomics™) allows for additional separation, accumulation and accurate quantification of peptides at a high sequencing speed of up to 120 Hz. Many immunopeptides exist primarily as a singly charged species. These peptides are readily discriminated from multiply charged ion signals by their Collisional Cross Section (CCS), reducing the interference usually occurring from co-isolations in m/z only approaches. This alone is leading to cleaner MS/MS spectra.

More than just a dual source instrument: Introducing new powerful tools on timsTOF fleX

Understanding disease often requires comprehensive knowledge about molecular processes at their spatial occurrence. Current approaches are not seldom limited by either the analytical depth, or the lack of spatial information. We will present a unique workflow, called SpatialOMx, which bridges this gap by providing deep Omics data that is connected to the tissue origin through MALDI Imaging. Integration of PASEF empowered 4D-LipidomicsTM results from tissue extracts make lipid annotations in MALDI images more reliable through CCS-matching, which will be demonstrated using a powerful software solution.

On the timsTOF fleX, the enabled MALDI-2 technology empowers your MALDI Imaging studies with unsurpassed sensitivity and ability to map more compounds. Toxtyper® - a complete solution for rapid and comprehensive forensic toxicology screening

Forensic analysis is challenged by the continuous emergence of new psychoactive substances (NPS), requiring the development of comprehensive and specific screening methods. The Toxtyper solution provides screening and confirmation in one analysis and can be used as a toxicology ”push button solution” with ready to use LC-MSn methods. Toxtyper’s screening workflows cover drugs and drugs of abuse (DOA) in a variety of samples including saliva, serum, plasma and urine.

Reliable identifications are based on a comprehensive full scan MSn library search. The manually curated Toxtyper spectral library contains nearly 1200 compounds. In addition, specific smaller sub libraries are available for synthetic cannabinoids and psychotropic drugs, or the large Maurer/Wissenbach/Weber library holding 6500 drugs and their metabolites for a comprehensive urine screening.
