Overview

Description:

The FT-IR/Raman Training Course is designed for those users new to data acquisition and sample preparation. Laboratory teaching emphasizes measurement using various sampling accessories and spectral manipulations with the OPUS software. Three optional special topics are given on Thursday on specific advanced techniques.

The course will consist of lectures, laboratories sessions, and demonstrations which emphasize basic principles of FT-IR instrumentation, data processing, sampling techniques, the OPUS/IR PC software, and introductory instrument maintenance and troubleshooting.

Prerequisite:

A working knowledge of the windows 10 operating system operating system and a familiarity with your Bruker FT-IR and FT-Raman instrument. The basics aspects of the operating system (e.g. copying files, formatting disks, installing printers) can be practiced in the tutorials provided with the operating system.

Please bring your laptop with OPUS software installed if available.

March and December course takes place virtually. June and September courses take place in person. All are held in two two-hour sessions each day, from 9 a.m. to 11 a.m. and from 1 p.m. to 3 p.m.

Monday:

1)  Introduction to FTIR spectroscopy:

  • Molecular vibration and light absorption
  • Interferometer: constructive and destructive interference
  • Interferogram of monochromatic, polychromatic and broadband sources
  • Advantages of FTIR spectroscopy
  • Principles of data processing: folding, apodization, zerofilling; phase errors
  • Spectral resolution

2)  Optical components of FTIR spectrometer:

  • Sources
  • Beamsplitters
  • Detectors
  • Windows
  • Mirrors
  • Polarizers
  • Optical filters

3)   FTIR Sampling Techniques:

  • Transmission
  • Reflectance
  • DRIFT
  • ATR

4)   Spectral Interpretation:

  • Alkanes, phenols, ethers, ketones, acids, etc.

5)   FTIR test protocols:

  • Validation test setup
  • Running OVP tests

6)   Instrument care:

  • Basic maintenance and troubleshooting
  • Optical components exchange

1)   Introduction to Raman:

  • Infrared vs. Raman spectroscopy
  • Dependency of the excitation line
  • Spike correction
  • Fluorescence and thermal background

2)   OPUS I: Basic functions and data acquisition:

  • Software overview
  • File structure
  • Working with files
  • Setting experimental parameters

3)   Lab session I, data acquisition:

  • Effect of spectral resolution
  • Effect of apodization function
  • Effect of zerofilling
  • Effect of number of scans and S/N evaluation

4)   OPUS II: Data manipulation and evaluation:

  • Base line correction
  • Spectral subtraction
  • Library search
  • Plotting spectra
  • Quantitative analysis

5)   Lab session II, data manipulation:

  • Exporting data
  • Peak picking
  • Plotting with peaks
  • Baseline correction

  • 3D file structure
  • Chemical image

4)  Quantitative analysis:

  • PLS data modelling

1)   Rapid scan and step-scan time-resolved spectroscopy:

  • Principles of rapid- and step scan FTIR
