Superior Imaging for Advanced SMLM and Sub-Cellular Omics Applications

Bruker's Vutara VXL is a localization microscopy platform with two modes of operation. Standard super-resolution single-molecule localization microscopy (SMLM) mode uses a unique bi-plane detection with measured point-spread function (PSF) that enables superior 3D detection far off the coverslip (typically used for dSTORM, PALM, and DNA-PAINT). The large field-of-view (FOV) mode is primarily used for sub-cellular omics applications, such as chromatin tracing and smFISH. We will discuss the optical and software implementation of the two methods and their benefit for various applications.

The overlap of excitation and emission spectra limits the number of probes a microscope can distinguish in standard light microscopy and SMLM implementations. Bruker offers a fluidics platform for the Vutara VXL microscope that combines fluidics hardware with deep integration into the SRX software. We will discuss how this platform dramatically increases the number of probes for DNA-PAINT and chromatin tracing experiments.
