The analytical advantages of matrix assisted laser desorption ionization (MALDI) TOF MS have been demonstrated for more than 25 years. MALDI-TOF Technology

Over the last 25 years, matrix-assisted laser desorption/ionization (MALDI) mass spectrometry has evolved into a powerful, highly versatile technology with a diverse range of applications. MALDI coupled to time-of-flight mass spectrometry (MALDI-TOF MS) is used to sequence proteins, map biomolecules in tissues, identify microorganisms, and analyze several thousand biochemical assays in a day.

 

The sample for analysis by MALDI-TOF MS is prepared by mixing or coating with a solution of an energy-absorbent matrix which entraps and co-crystallizes the sample when dried. The matrix is ionized with a laser beam, and transfers the charge to the analytes, generating singly charged ions from analytes in the sample that are then accelerated. Ions are separated from each other on the basis of their mass-to-charge ratio (m/z) before being detected and measured using the TOF mass analyzer.

 

The high throughput and speed associated with MALDI-TOF MS has made this technology an important tool for large-scale proteomics, imaging studies, and other analyses.

MALDI Applications

Since the discovery of MALDI in the mid 80's this fast and high-throughput enabling ionization made its way into a variety of applications. The inherent speed of MALDI-TOF analysis from sample to result allows for fast answers to your analytical questions. The laser aiming at different positions gives rise to spatial, molecular information as used e.g. for MALDI Imaging for tissues.

MALDI Imaging

For MALDI Imaging, rapid data collection speeds, maintenance of critical spatial resolution, and broad dynamic range are mandatory, and, in conjunction with market-leading SCiLS™ Lab software, are addressed best using ultrafleXtreme or rapifleX® instruments. The unbeaten speed of the rapifleX® enables ultra-high throughput screening and together with our MALDI PharmaPulse 2.2 solution rapid delivery of needed results is ensured. These data are also used to classify tissue – so called tissue typing.

Polymer Analysis

MALDI-TOF technology offers a unique depth of characterization for many diverse polymer classes. Including bulk material screens, pharmaceutical development, or finished surface analyses, MALDI-TOF MS offers direct molecular weight distributions in fast and versatile workflows.

Quality Control

With a variety of available ionization matrices and the option for analysis in both positive and negative ion modes, MALDI-TOF MS and MS/MS workflows offer critical advantages in the quality control of peptides, oligonucleotides and many classes of polymers. Depending on the analytical depth needed microflex® LRF or autoflex® maX series in combination with BioPharma Compass® software are perfect solutions.

Biomarker Discovery

Molecular changes originate at the cellular level and solution-based techniques can lack sensitivity to capture changes produced from small cell populations, whereas MALDI can work directly of tissue.

Proteomics

Protein or peptide characterization using MALDI-TOF MS workflows quickly provide primary protein sequences of intact protein targets in just minutes. Also, higher level protein structure can be examined via the detection and localization of di- and trisulfide bonds. ultrafleXtreme and rapifleX® are the most suited MALDI-TOF instruments to perform top-down, middle-down, and bottom-up sequencing approaches as well as glycoprotein analysis.
